A new DNA detection
system could finally deliver
on one of the longtime
promises of genetic
medicine: on-the-spot
diagnosis of disease.
If successful, the result
would be a handheld device
that not only provides
instant diagnosis, but also
detects evidence of
bioterrorism in minutes
rather than hours. And do it
a lot more accurately than
current technology.
It was developed by
researchers at Northwestern
University, who have been
working on developing better
detection systems for DNA
for the past decade. By
incorporating nanotechnology
and electrodes, they say
they've finally come up with
a small, instantaneous and
extremely accurate method of
identifying DNA.
"This one will likely fit
the major need for a
handheld device that can
diagnose genetic and
infectious diseases," said
team leader Dr. Chad Mirkin,
who is director of
Northwestern's Institute for
Nanotechnology.
The fantasy tool for
biological warfare
detection, meanwhile, has
been a smoke detector-like
device. But the required
technology has lagged, until
now.
A privately funded company,
Nanosphere, in Northbrook,
Illinois will commercialize
the technology.
A prototype for genetic
research purposes will be
available as early as the
end of this year, said
Vijaya Vasista, chief
operating officer at
Nanosphere. A device for
on-site diagnosis could be
available in three years.
Nanosphere has competition,
but none with so much
published evidence
supporting their technology.
The Northwestern researchers
have published four papers
on the technology in Science
and Nature, the top science
journals.
A Danish company called
Cantion is developing a
biochip device for medical
diagnostics and research
they say will cost only $50.
Thorion Diagnostics, another
nanotechnology spinoff
located next door to Cantion,
is also developing a biochip
device for doctors' offices
that will cost around
$5,000.
Meanwhile, Abbott
Laboratories and American
Home Products are testing
Cantion chips for developing
diagnostic assays, and
Faltum predicts that human
clinical trials will begin
within two years. Because
the chips are diagnostic,
FDA review would be quicker
than for a drug.
Polymerase chain reaction,
or PCR, is the technology
currently used to identify
DNA. Using PCR, scientists
replicate tiny amounts of
DNA to make a sample large
enough for analysis.
That process takes at least
an hour, and it is prone to
false positives and
negatives.
Using nanotechnology instead
of microscopic technology
greatly increases speed and
accuracy. The new technology
works 10 times faster and is
100,000 times more accurate
than PCR, the researchers
said. Details of the
research will be published
in the Feb. 22 issue of
Science.
The researchers tested the
technology on a synthetic
form of anthrax and it was
able to accurately and
quickly identify the
contagion, using a
combination of gold, silver,
"nanopores" and electrodes.
It works by analyzing the
rungs of the DNA ladder --
or double helix -- which are
connected by nucleotides
represented by the letters
A, C, G and T. The letters
stand for adenine, thymine,
cytosine and guanine.
A always connects to T, and
G always connects with C --
except when there are
problems. To find mismatched
pairs, or to find a sequence
of the letters that makes up
a specific disease or
infectious material,
researchers look at the
sequence of these letters
and how they're paired off.
In the new technology,
scientists separate strands
of DNA and place them in a
solution with nanoparticle
probes. The probes are
covered with other DNA
strands that will match and
bind to the strands being
tested if they are, for
example, DNA from an agent
such as anthrax.
"It's like a chemically
specific Velcro ball,"
Mirkin said. "It latches
onto whatever it's supposed
to latch on to."
Once the DNA binds,
researchers need to amplify
it in some way so they can
read it. The Northwestern
researchers used a modified
photographic developing
solution to amplify the gold
probes, which are then
covered with silver, which
conducts an electric
current.
The entire package then
fills up the space between
two electrodes and emits an
electrical signal if a
disease DNA or pathogenic
agent is present.
In PCR, researchers make the
DNA fluorescent, then view
it with a confocal
microscope that costs about
$60,000. The Northwestern
researchers eliminate this
step by using the electrodes
instead of fluorescence.
They also discovered that a
salt solution will break
apart any mismatched DNA
pairs, which was the
clincher for giving this
technique its extremely high
level of specificity:
100,000 to 1 rather than 2.5
to 1 with PCR.
The salt wash doubles as a
cooling mechanism for the
technology, eliminating the
extra step of temperature
control -- always a vexing
issue in any kind of chip
design.
Although Mirkin didn't state
a price for the device, he
said it would be much
cheaper than PCR.
"Look at how cheap silicon
chip technology is now," he
said. "We hope to
mass-produce (the
technology) and make it very
sophisticated for really low
cost."
In addition to detecting
disease or biological
warfare, the researchers say
the technology is so
sensitive that it could also
identify tiny but vital
genetic variations among
individuals called single
nucleotide polymorphisms, or
SNPs (pronounced snips).
Of the roughly 3 billion
nucleotides (A, C, T and G
pairs) 99.9 percent are the
same for everyone. The SNPs
that occur in the remaining
0.1 percent account for the
differences among people.
The colon cancer gene
variation, for example, is
an SNP. So the variations
can mark predisposition to a
disease, or determine how an
individual will react to
certain drugs.
The most difficult
nucleotide mismatch to
detect is when G binds with
T instead of C, a phenomenon
known by scientists as G:T
wobble.
"We chose a system that is
almost impossible to
differentiate," Mirkin said.
"But it was very easy to
differentiate with the new
